Abstract
Evolu&onary diversifica&on of RFX family genes as master regulators of ciliogenesis

Regulatory factor X-box binding(RFX) family have well conserved and specialized winged-helix DNA binding domain.
Previous studies reported that RFX gene family broadly interact with cilia genes. RFX2 stabilizes the binding of Foxj1
to the mulG-cilia gene promoter, to specifically promotes mulG-ciliated cell differenGaGon. RFX3 regulates
ciliogenesis in embryonic node, pancreaGc endocrine cells and moGle cilia in brain. Also, RFX7 regulates neuronal
ciliogenesis by regulaGng the expression level of RFX4. In situ hybridizaGon and Gssue expression data of Xenopus
embryos revealed that some RFX genes are expressed in epidermis, more specifically mulG-ciliated cells. In this study,
we idenGfied the transcripGonal target of RFX by performing Chip-seq analysis. We are also planning to idenGfy
deferenGally regulated ciliary genes for three types of cilia formaGons. To this end, we selected several RFX genes
and will discover RFX target genes involved in primary cilia, nodal cilia and mulG-cilia formtaion using Xenopus model.
