dc.description.abstract |
Intracellular Ca 2 is known to modulate adipocyte lipid metabolism as well as adipogenesis However, the regulation mechanism of intracellular Ca 2 levels during adipogenesis remains elusive In the present study, we found that the expression of STIM 2 β, which is involved in the regulation of store operated Ca 2 entry, is upregulated and is involved in the regulation of intracellular Ca 2 levels during adipogenic differentiation Knock out of STIM 2 β in 3 T 3 L 1 cells showed increased intracellular Ca 2 levels and increased differentiation efficiency, with more lipid accumulation and rapid transcriptional activation of adipogenic genes, especially proliferator activator receptor γ 2 (PPARγ 2 In addition, more cells underwent G 1 arrest in the absence of STIM 2 β, implying that altered intracellular Ca 2 levels by STIM 2 β KO could be involved in the cell cycle regulation during adipogenesis We also discovered that STIM 2 β KO mice showed hypertrophic adipose tissue development Our study provide evidences for the role of STIM 2 β in the regulation of intracellular Ca 2 levels during adipogenesis, to modulate the number of differentiating cells and PPARγ 2 transcriptional regulation |
