dc.description.abstract | Heterocyclic amines (HCAs) are generally formed during the cooking process of meat, poultry, fish, and other foods at high temperatures. Among various types of HCAs, 10 HCAs were classified as Group 2A and 2B carcinogens by IARC. However, there are few studies about monitoring of HCAs in agricultural products and their processed foods. In this study, for optimizing an analytical method, blank samples spiked with HCAs were mixed with NaOH and extracted with acetonitrile and methanol (MeOH). For purification of the samples, Chem-Elut cartridges (Agilent) and HLB cartridges (Oasis) were used, and each cartridge was eluted with dichloromethane/ethyl acetate and MeOH/acetic acid, respectively. Final extracts were analyzed using UPLC-MS/MS for determining HCA compounds. Mean recoveries of HCAs for blank samples were ranged from 83.5 to 111.8%. In addition, pomegranate juice (non-fat liquid), rice soup (non-fat solid), soybean oil (fat liquid), and margarine (fat solid) were regarded as representative samples of agricultural products. In these results, matrix-matched calibration curves of four sample types showed good linearity (r2>0.99). Method detection limits (MDLs) of these samples ranged from 0.034 to 0.062 ng/g, 0.011 to 0.103 ng/g, 0.014 to 0.150 ng/g 0.007 to 0.043 ng/g, respectively. On the basis of this study, we will plan to analyze HCAs for 351 agricultural products in South Korea. | - |
