Targeted delivery systems using various type of nanoparticles can maximize synergistic effect and enhance their delivery efficacy. Especially bio-interface hybrid complexes between biomaterials and nanoparticle have been reported as a powerful platform for the delivery platform by overcoming the biological environment such as protein corona. And targeted delivery platform based on nanoscale metal-organic framework (NMOF) to make a novel delivery systems has made tremendous progress with significant attention of many scientific fields. Their magnificent porosity with tailorable compositions and easier surface chemistry make them one of the most promising drug carrier. Here, we intend to advance the possibilities of MOF NPs one step further. We find that the intrinsic coordination bonding between the empty metal node site of MOF and many carboxyl and amine groups in protein surface is occured without significant deformation of protein but oriented conformation. Once thermodynamically stabilized in preferred orientations on the nanoparticles, it can be used as an antifouling layer while being present as a hard protein layer which can prevent blood serum protein absorption onto nanoparticles. Based on our findings, we engineered a “Plug & Play” drug delivery system using three types of antibody proteins (Anti-CD44, EGFR, and HER2) and MOF (MOF-808) NPs. This study provides insight into the reaction of MOF and bio interface for further variety of applications. |
